The influenza A virus hemagglutinin (HA) is a glycoprotein composed of two subunits. The HA1 domain is responsible for the receptor binding whereas the HA2 domain is responsible for fusion of the viral and cell membranes. HA has also been implicated in virulence and tissue tropism of the virus. The HA surface glycoproteins of influenza A viruses are known to be immunodominant antigens and are able to induce protective antibodies following vaccination and challenge. The present study attempted to identify an epitope on HA that could elicit protective antibodies. We screened two known neutralizing monoclonal antibodies (mAb) (CL194.9 and CA04) and a panel of 27 other known neutralizing and poorly neutralizing mAbs for binding activity to a synthetic peptide comprising residues 58-75 of the HA subunit II (HA2). Using ELISA and Western blot analysis, we screened the antibody binding to synthetic peptides that represent a well-defined linear epitope within HA2. We also performed a vaccinia virus recombinant (VVr) virus expressing the same peptide as a positive control for immunization. Our data indicate that mAb CL194.9 and CA04 bound to peptides 58-69, 61-69 and 67-74. All the mAbs that bound the peptides were able to neutralize virus infectivity in vitro; however, only CL194.9 and CA04 were able to protect the challenged mice.
